چکیده

There is a unique relationship between proline hydroxylation and collagen biosynthesis, since proline, rather than free hydroxyproline, appears to be the major precursor of collagen hydroxyproline (1, 2). The early stages of collagen biosynthesis have been shown to be similar to the steps involved in the biosynthesis of other proteins in that hydroxyproline with the highest specific activity appears in microsomal protein when proline-‘% is introduced into systems in vitro or in vivo (3, 4). It has been possible to obtain a cell-free system from whole chick embryos which converts proline-‘% to hydroxyproline-rC found in a microsomal protein (5, 6). Although the microsomal protein is obviously not in the fibrous form usual for collagen, it has many of the physical and chemical properties of collagen, as will be shown in this report. For purposes of convenience and clarity, the term collagen will be used in referring to the hydroxyproline-containing microsomal protein. Some properties and requirements of the cell-free system, consisting of microsomal and supernatant fractions, are described, and studies on the mechanism and site of proline hydroxylation are reported.

Conversion of Proline-14c to Collagen Trans-3-hydroxyproline-14c in the Chick Embryo.

trans-3-Hydroxy-nn-proline-3,4-3H was synthesized by reduction of N-carbobenzoxy-3-keto-DLproline methyl ester with NaBH4 (4). Uniformly labeled n-proline-r4C was obtained from New England Nuclear Corporation. Uniformly labeled 4-hydroxy-r.-prolineJ4C was generously donated to us by Dr. Chozo Mitoma. Seven-day-old chick embryos were obtained from the Duckworth Hatchery (Elkridge, Maryland) and ...

Collagen Metabolism in Osteolathyrism in Chick Embryos: Site of Action of Β-aminopropionitrile

Chick embryos were sacrificed at intervals after simultaneous injection of BAPN and proline-C(14), the collagen separated into neutral salt-extractable and residue fractions, and total hydroxyproline and hydroxyproline specific radioactivity determined in each fraction. Extractable collagen, measured as hydroxyproline, increased markedly and had a higher specific activity in BAPN-treated embryo...

Effect of Adenosine Agonists on the Proliferation and Differentiation of Chick Embryo Fibroblasts in Three Dimensional Reconstituted Tissue Constructs

Previous studies indicate that organ fibroblasts play an important role in wound healing, collagen production, remodeling processes and pathogenesis of progressive heart, lung, renal and hepatic fibrotic diseases. Several studies suggest a possible inhibitory role for adenosine in the regulation of fibroblast proliferation. The effect of adenosine A2 agonists on proliferation and differentiatio...

Studies in vivo on the biosynthesis of collagen and elastin in ascorbic acid-deficient guinea pigs.

1. After the administration of labelled proline to guinea pigs deprived of ascorbic acid for 15 days, the dorsal skin was examined 5 days later in an attempt to detect the presence of hydroxyproline-deficient collagen (protocollagen). The extent of incorporation of proline into skin collagens indicated a severe impairment of collagen synthesis. 2. A comparison of proline and hydroxyproline spec...
